 SYNTHESIS OF CHALCONES

PROCEDURE OF CHALCONE SYNTHESIS:

➢ In a solution of ethanol (EtOH) (10 mL), 2-hydroxy acetophenone (1.2

eq), the selected aldehyde (1 eq), and 40% NaOH were stirred at 65℃

and the progress of the reaction was monitored by TLC.

➢ The solvent was evaporated using a rotary evaporator.

➢ Upon completion, the excess of NaOH was neutralized by the addition

of HCl (2 M) and cold water, and the solution was neutralized, checked

using a pH paper.

➢ Solvent extraction was done using ethyl acetate (EtOAc) and water. The

organic layer and water layer were separated. The water solution was

washed again with ethyl acetate and brine to collect the remaining

product that was in the water layer.

➢ Then the organic layer was dried with Na2SO4 and filtered.

➢ The solvent was evaporated under reduced pressure.

➢ The crude product was purified by the help of Column

Chromatography using EtOAc & Hexane as mobile phase and 60-120

mesh silica as the stationary phase.

➢ The spots were separated and the solvent was evaporated.
